Resveratrol And Hair Loss Evidence
A newly released, 2012 study showed a cause and effect relationship between abnormally high levels of PGD2 and male pattern baldness. With topical application they found PGD2 prevents hair growth, and mice who were genetically predisposed to produce higher levels of PGD2 had diminished hair growth. They additionally discovered PGD2 levels were much elevated in balding scalp versus non-balding scalp tissue. The study concluded that one of the receptors involved in production of PGD2, GPR44, constituted a precise therapeutic target for hormonal hair loss in both men and women.

Abstract
Testosterone is necessary for the development of male pattern baldness, known as androgenetic alopecia (AGA); yet, the mechanisms for decreased hair growth in this disorder are unclear. We show that prostaglandin D(2) synthase (PTGDS) is elevated at the mRNA and protein levels in bald scalp compared to haired scalp of men with AGA. The product of PTGDS enzyme activity, prostaglandin D(2) (PGD(2)), is similarly elevated in bald scalp. During normal follicle cycling in mice, Ptgds and PGD(2) levels increase immediately preceding the regression phase, suggesting an inhibitory effect on hair growth. We show that PGD(2) inhibits hair growth in explanted human hair follicles and when applied topically to mice. Hair growth inhibition requires the PGD(2) receptor G protein (heterotrimeric guanine nucleotide)-coupled receptor 44 (GPR44), but not the PGD(2) receptor 1 (PTGDR). Furthermore, we find that a transgenic mouse, K14-Ptgs2, which targets prostaglandin-endoperoxide synthase 2 expression to the skin, demonstrates elevated levels of PGD(2) in the skin and develops alopecia, follicular miniaturization, and sebaceous gland hyperplasia, which are all hallmarks of human AGA. These results define PGD(2) as an inhibitor of hair growth in AGA and suggest the PGD(2)-GPR44 pathway as a potential target for treatment.
Because PGD2's correlation with asthma is well established, several drugs that down regulate PGD2 are already in clinical trials. PGD2 inhibitors would serve to promote hair growth. Scientists involved in this study, including Dr. Cotsarelis of stem cell research renown, state that they've already formulated pills containing the vital PGD2 blocking properties. But why wait for an expensive, patented medication. Resveratrol has been shown to significantly suppress PGD2 at low concentrations readily obtainable through oral dosing, as evidenced by the abstract presented below. Oral resveratrol has also been shown to have anti-asthmatic effects superior to that of dexamethasone, the side effect wrought, cortico-steroid of choice for managing asthma symptoms.

Abstract
Resveratrol is a natural phytoalexin occurring in grapes, vines and peanuts and possesses both antitumor and antioxidation capabilities. Its chemoprotective actions are attributed partially to its anti-inflammation effect. The present study is aimed at checking the inhibitory actions of resveratrol on the release of mediators from bone marrow-derived mouse mast cells (BMMC) in vitro. Mast cells were prepared by isolating bone marrow cells from intact mice femora and culturing them for 4 weeks in the presence of IL-3 and FCS. The release reaction was triggered by IgE or calcium ionophore (A23187). Mediated by IgE, the release of histamine and tumor necrosis factor-alpha was significantly inhibited by resveratrol at a concentration of 100 microM; IgE-mediated release of leukotrienes and prostaglandin D (2) was also strongly suppressed at concentrations of 100 and 10 microM.Also, A23187-mediated release of histamine and leukotrienes release was strongly reduced by resveratrol at concentrations of 100 and 10 microM, respectively. Resveratrol exhibited its behavior without a significant cytotoxic activity against mast cells. In conclusion, resveratrol is a potent non-selective inhibitor of mediator release from activated mast cells and deserves further investigation of its biological modulations.
The ideal dose of Resveratrol has been a subject of debate for some time. The scientific community is coming to some agreement, that a dose between 200-300 mg optimizes health benefits for human use. Possible New Hair Loss Cure
"Triarco”, a Bio-Tech company based in New Jersey, released some very intriguing results of a pilot study for their patented prostate product Alphastat®, which inhibits 5-alpha reductase. The three-month study on male patients ages 55-68 reported a 40% decrease in blood levels of DHT, which has some obvious implications for treating hair loss.
In addition to improved Urogenital symptoms, there were, unlike Proscar and Avodart, reports of increased, not decreaesd sexual function.
As you know, pharmaceutical 5 alpha reductase inhibitors prevent balding and promote hair growth by lowering serum DHT. The documented side effects of pharmaceutical 5 alpha reductase inhibitors include sexual dysfunction, reduced libido, and depression. There were evidently no reports of any negative side effects in this study.
Triarco stated “We are very pleased with the results of this clinical study because it confirms the results of the pre-clinical studies and is, to our knowledge, the first natural product shown to lower PSA and serum DHT levels in humans."

Study Summation
The investigators gave 40 healthy male subjects between the ages 40 and 70, 800 mg or 2g of Alphastat, for two weeks. Below you can evaluate the effect of both dosages on the concentration of serum testosterone in the blood of the test subjects.
More importantly for hair growth, both dosages rapidly reduced serum DHT, as noted below.
Estradiol level was reduced as well, with the smaller dose being slightly more effective than the larger dose (see below).
The study appears to be scientifically valid, with a sufficient sample size of 42 males, measurable blood tests, and sound statistical methodology. It was published in a peer-reviewed scientific journal. This means that at least two independent researchers have reviewed the article and deemed it to be fit for publication
Comment
There is no evidence suggesting Saw Palmetto per se, has any effect whatsoever on Testosterone levels or serum DHT levels (Saw Palmetto *does* moderately inhibit DHT binding to receptors). The desirable effects on hormone blood levels produced in this study are due to Astaxanthin.
Astaxanthin is also documented to reduce bodyfat, increase stamina, and maintain glycogen stores in animal models.
Dissecting Alphastat Ingredients
Alphastat® is comprised of 200 mg of a “patented complex” of Saw Palmetto Extract extract and Haematococcus pluvialis algae extract, providing free fatty acids and Astaxanthin.
Saw Palmetto Extract is typically used at about 300 mg, so it is likely that most of this complex is Saw Palmetto Extract.
Astaxanthin is a relatively expensive supplement and is generally available in 2 to 4 mg capsules.

“This is an impressive difference,” says Dr. Glatt. “He looks 10 years younger!” Um, no, he looks like a scary robot! A little botox is maybe one thing, but our beloved Everett is totally unrecognizable! In fact, he looks kind of like Bruce Jenner, another awful surgery victim.
We can’t help but notice a surge in plastic faces amongst Hollywood men like John Travolta, Ray Liotta and Mickey Rourke. Nicholas Cage’s hair plugs also make us shudder. Why not go the route of Bruce Willis and shave it all off and make that your look? No one thinks you miraculously grew hair after balding for 20 years. The plugs are worse than a bad toupee.

Resveratrol and Curcumin For Hairloss
I will say that in a lot of these hair loss forums there's a lot of buzz about how Curcumin enhances the effects of Resveratrol so of course, I'm getting in on this one.
About 2 months ago went to the health food store and bought some curcumin and a bottle of resveratrol but I didn't really know what I was doing.

Rogaine on Steroid to Stop Hair Loss
You know years ago I glopped a bunch of Rogaine on my head to try and grow some peach fuzz on the top of my head because the FDA said I could.
Now anyone suffering from male pattern baldness or female hair loss isn't looking for peach fuzz. We want hair and Rogaine typically was not the answer.

From a point of view of molecular biology, the presence of a vanilloid receptor on the cell membrane and close to the mitochondrial structures can explain the induction of apoptosis or planned death of cells that would have become neoplastic. Moreover, in the Central Nervous System there are various receptors of capsaicinoids and this could explain the enhancement of the anti-aging action of resveratrol by curcumin.
The group of patients that has assumed only resveratrol didn't show any modification on the PSA, while the group treated with resveratrol and curcumin showed with great surprise a reduction, sometimes also relevant, in variable time periods. Moreover, during this testing, the patients treated with resveratrol and curcumin surprisingly showed positive improvements in the treatment of psoriasis and the attenuation and even a stop of the hair loss, a reduction of the hair graying and the rebirth of hair in bald areas sometimes even of the original color too.
Regarding its action on hair growth, even if not yet known, it is probably linked to the release of P substance. The growth and the pigmentation of the hair follicle would depend on mesenchymal-epithelium-neiroderic interactions not very well-known yet, where the P substance would stimulate the hair growth in vivo (RaIf Paus et al Investigation Labs, VoI 71, No 1 , p.135, 1994).

Another object of the present invention is a combination comprising resveratrol and curcumin for the simultaneous, separate or sequential use in the treatment of cancer, as for example, the prostate carcinoma, in the treatment of skin diseases as psoriasis and for the prevention and the treatment of alopecia.
The invention in fact can vary in its dosages and in its components, adding other components similar to the one suggested, but without altering the conceptual basis of the invention.
In this testing, apart from a relevant anti-cancer action on hormone-insensible prostate neoplasia, an effect on some patients has also been the stop of the hair loss and the growth of hair, sometimes of the original color too. Therefore patients without any neoplastic disease, but affected by alopecia or hair loss, have been included to the testing.
MATERIALS AND METHODS
The group treated only with resveratrol didn't show any relevant variation, except from two cases that showed a slight decrease.
Capsures of 4 pills a day let to a reduction in the level of PSA after 60 days to 7.4 and then to 6.80. The patient suffered also of psoriasis of the scalp, that disappeared after the treatment and that by now, after 8 months has not reappeared. The compound containing just resveratrol as active agent (Resverage) didn't cause a reduction of the PSA value and this can be due to the low bioavailability of the molecule. In the cases of prostate carcinoma the efficacy of the association of resveratrol with curcumin can be due to a higher concentration in the circulation and therefore in the tissues explained by the lipophilia of the active elements of curcumin.
The association of resveratrol and curcumin seems to have a synergic effect that enhances the anti-inflammatory power of the two active elements. It is an essential activity because in the prostate carcinoma there is a phlogistic action caused by hyperincretion of enzymes as the cyclooxygenase 1 and 2.
As far as the action on hair is concerned, it is unquestionably due to the administration of the two combined active elements, since in the compound with just resveratrol no result has been shown and in the many cases in India and in Europe where Curcumin has been employed, any effect on hair has never been reported. The most important aspect rarely highlighted is that curcuminoids are capsaicinoid substances.
Those fibers, whose nerve cell is in the posterior roots, once stimulated release neuropeptides like the P substance, Neurokinin A and the Calcitonin Gene Related Peptide (CGRP). Experimental researches demonstrated that the desensibilization of those fibers leads to skin lesions localized in the cervical region of the rat. These experiments indicate the existence of a tropic function. It is probable that the loss of hair is a result of the deficit of neuropeptides released by the fibers that innervate the scalp.
Therefore it is possible that the administration of a substance that has as activity similar to capsaicin, as the Curcumin that metabolizes like an active curcumoid, can stimulate the sensorial fibers that have a tropic function, and that resveratrol has a vehicular function on the tissues. Therefore, the composition of the present invention is active on cases of hormone-insensible prostate carcinoma but in particular on hair, as our research has proven. The efficacy of the composition according to the present invention also depends on the great lyophilic properties of a capsaicinoid, curcumin on the anti-inflammatory effect of the two substances.
The interference of the hormonal action on hair loss is unquestionable, even if capsaicinoid-sensible sensory innervations are an ancestral phenomenon that does not depend on the hormonal mechanisms, but through a metabolic way different from the ordinary one. The growth and the pigmentation of the hair follicle seem to be controlled by epitelius-mesenchimoneuroechtodermmic interactions not very well known, but with an important role of the P substance on the hair growth in vivo.
